Joel Miller (Troy Baker) is the protagonist of The Last of Us. Originally from Texas, Joel was a single father in his late 20s when the initial Cordyceps outbreak occurred.

Ellie

Ellie (Ashley Johnson) is a brash, precocious 14-year-old girl, orphaned at a very young age, with no recollection of how the world was like before the collapse of modern society. Despite having grown up in the hard, unforgiving post-apocalyptic world of The Last of Us, her rough, confrontational nature is tempered with a penchant for lighthearted humor, a strong desire for emotional connections, an existential fear of being alone, and a genuine childlike curiosity for the world outside the quarantine zone and before the infection.

Marlene

Marlene is the leader of the Fireflies, a resistance group fighting against the oppressive government regime in The Last of Us. She has an easy charisma and although she does have her moments of self doubt, her strong belief in the righteousness of her cause always pulls through.

She has a special relationship with Ellie, having known the girl's mother and having been personally tasked by the mother to look after Ellie. Marlene is seen as both mentor and older sister by Ellie.

David

David is the leader of a community of survivors in The Last of Us. He speaks in a rational manner, hiding a darkness to Ellie during their first interactions that the post-apocalyptic world has brought out of many other survivors of his ilk.

He is introduced in the Winter chapter, stumbling upon Ellie after she had killed a deer. He offers medicine from his camp to trade with her meat. Ellie forms an uneasy alliance with him when they both fall under attack by a big group of Infected. His true colors are soon revealed after they survive the encounter.
